ANALISIS FILOGENETIK NIBUNG (Oncosperma tigillarium (Jack) Ridl.) “FLORA MASKOT” RIAU BERDASARKAN SEKUEN GEN rbcL DENGAN METODE MAXIMUM PARSIMONY (MP)
(perpustakaan UR, 2021-06) Hidayanti, Retno Esti; Fitmawati, Fitmawati
Taxonomic studies using morphological markers on nibung are difficult to do, because generative characters such as flowers and fruits as important characters are difficult to find in the out of season period. Thus, an alternative analysis using molecular methods is required. The study aimed to analyze kinship relationships and to reconstruct phylogenetic trees based on the character of the rbcL gene. DNA Extraction Procedure used CTAB method with modification. DNA fragments were sequenced at the First Base Laboratories Malaysia through PT. Genetika Science Indonesia followed by alignment using Bioedit application and phylogenetic analysis using Maximum Parsimony (MP) methods in PAUP* application version 4.0a169. The MP analysis grouped three collections of Oncosperma tigillarium into monophyletic clads with a bootstrap value of 100%. This grouping shows that the three collections of Oncosperma tigillarium from Bengkalis Island and Oncosperma tigillarium from Rupat Island have a close kinship based on rbcL genes

ANALISIS FILOGENETIK TIGA KULTIVAR SALAK DI DAERAH ACEH BERDASARKAN PENANDA rbcL MENGGUNAKAN METODE NEIGHBOR JOINING
(perpustakaan UR, 2021-06) Darmawan, Haura Zavira; Fitmawati, Fitmawati
Aceh is one of the center areas of wild type and cultivated snake fruit (salak) as it has a various topography. The massive deforestation resulted in reduced salak habitat and salak diversity in Aceh. In addition, lack of information about salak kinship in Aceh and difficulty in morphological-based taxonomic studies lead to the requirement of molecular approach which facilitate the study of salak taxonomy. This study aimed to reconstruct phylogenetic trees and analyze the kinship relationship of Salacca species in Aceh based on rbcL gene sequence using neighbor joining method. DNA extraction used CTAB method with modification. Phylogenetic analysis using neighbor joining method showed that Salacca sp. (NH.07) Lembah Seulawah had the longest evolutionary rate compared to other two salak samples.

ISOLASI DNA DAUN MANGGA KASTURI (Mangifera casturi K.) MENGGUNAKAN GENOMIC DNA MINI KIT (PLANT) GENEAID
(Elfitra, 2022-06) Rahmadani, Pidiyah; Fitmawati, Fitmawati
Mangifera casturi is an endemic mango from South Kalimantan that has become extinct in the wild and population tends to decrease. Information on the diversity of Mangifera casturi can be seen molecularly, considering that the Mangifera casturi is a plant that rarely appears so organs that are often found are incomplete for morphological analysis. Molecular analysis requires important steps, namely DNA isolation, because it will determine the quality and quantity of DNA to be used for further analysis. This study aimed to isolate the total DNA in 3 individual Kasturi mango plants with the help of a Genomic DNA plant minikit (Genaid). The isolation results showed that the total DNA of the Kasturi mango was successfully isolated using an isolation kit. The resulting total DNA has thick bands without smear.

(perpustakaan UR, 2021-07) Silvia, Mega; Fitmawati, Fitmawati
One of the largest mango diversity center in the world is Indonesia. Taxonomically, three types of Mangifera namely M. Laurina, M. indica and M. sumatrana require better proof to obtain more certainty of taxonomic status. The source of taxonomic evidence that can clarify the taxonomic status of the three types Mangifera is characters of pollen morphology because it is unique and pollen of different plants has a different shape, size and wall-type of pollen. This research was conducted from November 2020 to March 2021. Sampling was conducted around Pekanbaru and Kampar areas, Riau Province. The pollen observation was conducted in the Botanical Laboratory of the Department of Biology, Faculty of Mathematics and Natural Sciences, Riau University. Results of the study obtained 5 polen characters. Similar properties of morphological characteristics of pollen are monad polen unit, polar view angular, equatorial view circular/oval, polarity polen isopolar, while the differences were found in size and shape of the pollen. The character of the size and the shape of the pollen can contribute as one of the distinguishing characters among the three types of Mangifera

Musi Rawas Utara District (MURATARA) is a new autonomous region (DOB) in the South Sumatra Province. Deforestation in this region is very high to build regional facilities and infrastructure which has a negative impact on the occurrence of wild plants such as macang varieties (Mangifera foetida L.) before they are known its occurrence in nature. Therefore, it is necessary to characterize macang varieties to know the occurrence and can be conserve for germplasm source. The purpose of this research was to characterize macang varieties (Limus, Batu and Manis) in MURATARA District based on the morphological characters. This research was carried out using survey method. Morphological characters observed were stature of trees, leaves and stems. The results showed that there were variations in tree stature, part of leaves and stems between Limus, Batu and Manis varieties.

Sampling was carried out using a survey method. Samples were analyzed using the LC-MS system. Chromatogram was analyzed using the MassLynx program to obtain the compounds contained in the sample. Determination of compound names was based on the Chemspider database and compound groups based on PubChem. Grouping of the three origins of Sungkai was carried out using the NTSYSpc2.02 program. There were 296 metabolite compounds which were used for grouping sungkai from trhee regions. Results showed that ekstract leaf sungkai from three locations contain compounds belonging to the class of flavonoids, phenolics, alkaloids, amino acids, aromatic compounds, sesquiterpenoids, carboxylic acids, essential oils, medium-chain fatty acids and nucleic acid. Three sungkai from three locations were divided into two groups with similarity coefficient values ranging from 31% -48. Group I is from Kuantan Singingi and group II is from Pekanbaru and Siak. The metabolites found in this study are expected to be useful as basic data in the field of phytopharmaca and medicine.

OPTIMASI METODE ISOLASI DNA GENOM PADA TANAMAN NIBUNG (Oncosperma tigillarium (Jack) Ridl.)
    (2020-06) Novela, Deti; Fitmawati, Fitmawati
Nibung (Oncosperm tigillarium (Jack) Ridl.) belongs to family Arecaceae. Characterization based on morphological features in nibung is difficult, therefore molecular characterization is necessary to be done. Molecular characterization based on DNA markers provides a fairly high degree of accuracy. Plant DNA isolation is the key step in genetic and molecular experiments. One factor that has an important effect on DNA isolation is leaf condition. This research was carried out with the aim to find out the quality of DNA from the isolation of fresh nibung leaves and dried leaves storing in silica gel. Based on the results of this study, DNA isolation of the nibung dried leaves showed DNA band with good quality compared to fresh leaves.
