Rukam is one of the species of Flacourtiaceae which is widely distributed all over Sumatra. This study was aimed to isolated secondary metabolite compound from the leaf of rukam and toxicity of the compound was determined by BSLT method. A secondary metabolite compound (FR-H-03) has been isolated by flash chromatography. The compound was characterized by FT-IR spectroscopy, which showed the presence of the OH group, aliphatic C-H, methylene group (CH2), methyl (CH3), cyclic alkene C=C and C-O (alcohol), also the melting point was obtained at 260-262oC. Toxicity assay showed that the n-hexane extract and fraction were non-toxic with LC50 value >1000 ppm, and FR-H-03 compound was toxic with LC50 value 13 ppm.
