Item BIODEGRADASI POPOK BAYI MENGGUNAKAN JAMUR SELULOLITIK Trichoderma asperellum T.N.J63 DENGAN FERMENTASI PADAT(2016-05-10) Damayanti, Nelvi; Nugroho, Titania Tjandrawati; Haryani, YuliBaby diaper contents cellulose pulp that can be used as substrat in cellulose production. The objective of this study was to investigate baby diaper biodegradation abilities of cellulolytic fungi Trichoderma asperellum T.N.J63 which isolated from orange farmland in Riau province. Biodegradation process by solid fermentation was carried out for 10, 15, 20, 25, and 30 days. Parameters examined were activity of crude extract cellulose using Nelson-Somogyi method, organic carbon (TOC) using gravimetric method, nitrogen total (N-total) using Kjedahl method, and C/N ratio. The result showed that baby diaper can be useed as substrat to produce cellulose. The highest activity of crude extract cellulose was found at 25 days fermentation by T.N.J63 LBKURCC2 (5,78± 0,37) × 10-3U/mL. Total organic carbon was decreased during 30 days fermentation (29.668%), N-total during 30 days fermentation (0.418%), and C/N ratio was found to be 106.351.Item BIOSINTESIS NANOPARTIKEL PERAK MENGGUNAKAN EKSTRAK AIR RIMPANG JAHE MERAH (Zingiber officinale) DAN UJI AKTIVITAS ANTIBAKTERI(2016-10-19) Putri, Eka Marissa; Haryani, Yuli; Kartika, Ganis FiaThe study aims to biosynthesis of silver nanoparticles using extract of Zingiber officinale rhizome. This methode was performed by reducing AgNO3 at room temperature in 45 hours with secondary metabolites of extract rhizomes. The formation of silver nanoparticles was confirmed by doing characterization using UV-Vis spectrophotometer. The silver nanoparticles can be observed at 402, 404, 412, 411, 417 and 419 nm from the UV–vis spectrum for ratio 1:1; 1:3; 1:4; 1:5; 1:10 and 1:20 (exract: AgNO3). Antibacterial activity was performed against Staphylococcus epidermidis and Pseudomonas aeruginosa. The results showed that silver nanoparticles was actively inhibit the growth of Staphylococcus epidermidis. Microwave was used in order reaction time. Various ratio of Alpinia galanga rhizome rhizome aquoeous extract and 10-3 M silver nitrate solution (v/v) were 1:1; 1:5; 1:10 and 1:20 for the reaction time of 30; 60; 90; 120; 150 and 180 s. Color changing was to started at 60 s reaction and the color was darker by the increasing of incubation time. These biosynthesized silver nanoparticles were characterized by using UV-Vis spectrophotometer. UV-Vis spectra showed that the compounds were silver nanoparticles because they produced peaks at 405-500 wavelength. Among those various of ratio, it ws found that the ratio of 1:10 produced the best spectra at 120 s reaction time. AgNO3 10-3 M solution was reduced using the extract at room temperature by stirring for various ratio of extract and AgNO3 10-3 M solution. Color changing of the solution was detected at 9 hours reaction time. The color tends to be darker by the the increasing of reaction time. The formation of silver nanoparticles was confirmed by doing characterization using UV-Vis spectrophotometer. Nanoparticles were detected in the wavelength range of 430-450 nm. Antibacterial activity of silver nanoparticles produced was determined for the ratio of 1:5 at a reaction time of 19 hours for silver nanoparticles from Alpinia galanga. Antibacterial activity was performed against Staphylococcus epidermidis and Pseudomonas aeruginosa. The results showed that silver nanoparticles of Alpinia galanga was active against Pseudomonas aeruginosa. Lactococcus lactis ATCC11454 is a specific strain of lactic acid bacteria (BAL) frequently utilized in the food industry for its efficient production of nisin, a peptide known for its biopreservative properties in food products. The objective of this research is to evaluate the application of Cell-Free Supernatant (CFS) from Lactococcus lactis ATCC11454 on broiler chicken fillets concerning changes in pH values. Chicken fillets were coated with and without CFS from L. lactis ATCC11454, and then stored at two different temperatures. The percentage increase in pH values of fillets without CFS coating was higher compared to fillets with CFS coating.Item ISOLASI BAKTERI ENDOFIT DARI BRUGUIERA sp. A total of 47 isolates of endophytic bacteria were isolated from the roots of Bruguiera sp. It was found that 14 isolates were active in inhibiting the growth of Vibrio alginolyticus, 4 isolates were active to inhibit the growth of Escherichia coli, while no isolate gave any inhibition zone for Staphylococcus aureus.Item ISOLASI JAMUR ENDOFIT DARI AKAR MANGROVE KABUPATEN BENGKALIS(wahyu sari yeni, 2019-04-30) Juwita, Dita; Puspita, Fifi; Haryani, YuliMangroves strives under extreme conditions such as highly fluctuating salinities, extreme tide actions, strongwinds, high temperatures, muddy and anaerobic soils. This study aims to isolate endophytic fungi from the roots of mangrove. Endophytic fungi isolation was carried out by sterilizing samples using 70% alcohol, 5.25% NaOCl and sterile distilled water, then inoculated samples on PDA media using direct inoculation techniques and scatter plates. In this study, 28 isolates of endophytic fungi were isolated from mangrove root samples identified as Bruguiera sp. and Ceriops tagal (Perr.) C.B.Rob. Cellulase enzyme can be degrades of cellulosewaste containing that enable for the processing of waste to economically priced products such as glucose. In this research, optimization of the production of cellulase enzymes from isolates of S-16 and S-22 with a variety of media composition, temperature and concentration of carbon source production. The quantitative assay for enzyme activity was determined by Nelson Somogyi method with carboxymethyl of cellulose (CMC) as a substrate. The highest cellulase enzyme activity of S-16 obtained in a media production (KH2P04; MgS04.7H20; NaN03; CaC12; NaCl; CMC) of 88x10-3 U / mL, and the optimum concentration of source of carbon at 1% obtained for 50.36 xlO-3 U / mL, whereas of S-22 obtained in a media production (KH2P04; MgS04.7H20; FeS04.7H20; NaN03; KCI; CMC) of 14x10-3 U / mL, and the optimum concentration of sources of carbon at 1% obtained for 14 , 7x10-3 U / mL. Penggunaan bahan tambahan makanan tersebut bertujuan untuk membuat makanan yang diproduksi tampak lebih berkualitas, tahan lama, menarik, serta memiliki rasa dan tekstur yang sempurna. Namun yang menjadi permasalahan adalah ketika terjadi pemakaian bahan tambahan makanan yang tidak sesuai dengan peraturan yang ada dan tidak adanya pengawasan dari pihak terkait. Penggunaan bahan tambahan makanan pada industri saus tomat telah banyak dilaporkan. Diantaranya adalah penelitian yang dilakukan oleh Siaka 2009, yang menunjukkan kandungan asam benzoat dalam saus tomat yang melebihi kadar maksimum yang diperbolehkan menurut Permenkes No. 722/Menkes/Per/IX/1988. LBKURCC34 DENGAN VARIASI WAKTU INKUBASI(2020-08) Hardianti, Vinkha; Nurulita, Yuana; Haryani, YuliLiquid Chromatography Mass Spectrometry (LC-MS / MS) is an analytical technique that combines the physical separation capabilities of liquid chromatography with the specificity of mass spectrometry detection. This method can identify the differences of compound from the extracts. The ethyl acetate extracts produced from the co-culture of Penicillium sp. LBKURCC34 and Staphylococcus aureus with various incubation times (8 days, 11 days and 14 days) were identified with LC-MS. The results of secondary metabolites obtained showed that the peak of the dominant compound in extracts generally was at retention times ranging from 9-13 min. The data showed that the compound characteristics are more polar. Journal of Agricultural Technology 9(3):585-590. Researches on plant-derived endophytic fungi have grown in recent decades. Dahlia variabilis is one of natural products which potential as a source of antimicrobial agents. The purpose of this research was to isolate and characterize endophytic fungi from the tubers of Dahlia variabilis because derivation of bioactive compounds directly from the plants needs too many biomasses. Direct seeds method was used to isolate endophytic fungi from the samples. Samples were surface-sterilized by immersing in 70% ethanol, soaked in 15% peroxide acid, rinsed in sterile distilled water, and then put on the surface of Sabouraud-4% Dextrose Agar (SDA). Four isolates of endophytic fungi were isolated and characterized morphologically i.e., Monilia sp., Aureobasidium sp., Moniliella sp., and Sporothrix sp.Item UJI AKTIVITAS ANTIJAMUR DARI EKSTRAK JAMUR ENDOFIT Botryosphaeria rhodina TERHADAP JAMUR Candida albicans(Elfitra, 2023-04) Amalia, Fitri; Hendra, Rudi; Haryani, YuliEndophytic fungi are microorganisms that are able to form colonies in plant tissues without posing a threat to their hosts and that live in plant tissue systems at certain times. Botryosphaeria rhodina, isolated from mangroves in the Muara Sungai Siput region of the Bengkalis Regency, is one of the endophytic fungi with high biological activity. This study aims to isolate secondary metabolites from the extract of the endophytic fungus B. rhodina and assess their antifungal activity against Candida albicans. The microdilution method was used to determine the MIC (minimum inhibitory concentration) value, while the agar diffusion method was used to determine the MFC (minimum fungicidal concentration) value. Methanol and n-hexane extracts with concentrations of 1,000, 500 and 250 μg/mL were used as test samples. The results of the antifungal test revealed that the MIC values of the n-hexane extract, methanol extract, and MFC were 1,000 μg/mL, 500 μg/mL and 1,000 μg/mL, respectively, against C. albicans.Item UJI AKTIVITAS ANTIOKSIDAN EKSTRAK METANOL BUAH Solanum ferox L(wahyu sari yeni, 2019-01-15) Arief, Yohana; Haryani, Yuli; Yuharmen, YuharmenFree radicals atoms, molecules or compounds that have unpaired electrons, are very reactive and enstable. Cell damage, due to reactivity of radical compounds cause, it is of various diseases such as cancer, coronary heart disease, rheumatism, cataracts, and liver. In Bangladesh, decoction of plant Solanum ferox L is used to treat cough, asthma, fever, and vomit. Wheares, in India, it is as antirheumatic, antiviral, and anticancer. This study aims to determine the antioxidant activity of Solanum ferox L fruit by the DPPH method. Phytochemical preliminary test showed that young fruit contained alkaloid compounds, while ripe fruit contains flavonoids. Analysis results The antioxidant activity of the n-hexane extract of the acidic eggplant fruit showed very weak activity in young fruit compared to mature fruit because it had IC50> 500 μg / mLItem UJI ANTIBAKTERI EKSTRAK METANOL JAMUR Botryosphaeria rhodina DARI AKAR MANGROVE(Elfitra, 2023-04) Mawarni, Annisa; Hendra, Rudi; Haryani, YuliThe genus Botryosphaeria, which can flourish in a range of climatic conditions worldwide, includes the endophytic fungus Botryosphaeria rhodina. This genus's antimicrobial activity has been reported in several of its species. This investigation aims to assess the antibacterial activity of an endophytic fungus, B. rhodina, from the mangrove root, Xylocarpus granatum J. Koenig, against Gram-positive and Gramnegative bacteria, including Listeria monocytogenes ATCC 7644, Salmonella typhimurium ATCC 14028, and Bacillus subtillis ATCC 19659. With a clear zone diameter ranging from 16 to 22 mm, the results of the antibacterial test using the well diffusion method revealed that the methanol extract at a concentration of 500 μg/mL had antibacterial activity in the strong to very strong category for all pathogenic bacteria tested. The L. monocytogenes bacteria had the largest clear zone, measuring 22.03 mm by 0.30 mm.Item Uji bioaktivitas dan pemanfaatan fraksi aktif umbi, batang, bunga, dan daun dahlia (Dahlia variabilis) sebagai sumber bahan aktif obat penyakit kulit(2015-07-05) Haryani, Yulisebagai komoditas tanaman bias. Untuk meningkatkan pemanfaatannya, pada penelitian ini telah dilakukan isolasi dan pengujian bioaktivitas ekstrak dan fraksi tanaman dahlia. Ekstrak n-heksana dan metanol beserta fraksi-fraksinya dari umbi dahlia yang bunganya berwama merah memperlihatkan aktivitas yang baik dalam menghambat pertumbuhan Candida albicans, jamur patogen penyebab candidiasis. Pada konsentrasi 2%, fraksi ketiga ekstrak n-heksana dan fraksi kedua ekstrak metanol masih memperlihatkan aktivitas dalam menghambat pertumbuhan Candida albicans dengan diameter hambatan sebesar 15 mm dan 17 mm, berturut-turut. Ekstrak n-heksana dari batang dan daun dahlia yang bunganya berwama merah juga menunjukkan aktivitas terhadap Candida albicans. Di samping itu, juga dilakukan isolasi dan pengujian aktivitas antibakteri ekstrak etanol umbi, daun dan bunga tanaman dahlia yang bunganya berwama putih dan merah. Hasilnya memperlihatkan bahwa Uftibi, bunga, dan daun dahlia nierah mampu menghambat pertumbuhan Bacillus subtilis, sedangkan terhadap Escherichia coli hanya umbinya yang mempunyai aktivitas. Untuk dahlia putih, umbi dan daunnya mempunyai aktivitas terhadap kedua jamur uji, namun tidak untuk bunganya.Item Uji Kontaminasi (Samonella spp) Pada Jajanan Pinggir Jalan Di Sekitar Sekolah-Sekolah(2013-02-27) Haryani, YuliKontaminasi Salmonella spp. pada 14 sampel yang dibeli dari pedagang makanan dan minuman pinggir jalan di sekttar area sekolahan yang mewakili 7 lokasi berbeda telah dilakukan. Dari tiap lokasi yang diharapkan mewakili seluruh wilayah utama di Pekanbani dipilih 2 pedagang dari 2 sekolah yang beiteda secara acak. Dari 14 sampel yang diteliti, satu sampel (Teh es, Panam) positif tericontaminasi Salmonella spp. Dua sampel (Martabak dari Sukajadi dan Bakso dari Sail) tidak memberikan pertumbuhan koloni pada Xylose Lysine Deoxycholate (XLD) Agar, sedangkan 11 sampel lainnya memberikan pertumbuhan koloni pada Xylose Lysine Deoxycholate (XLD) Agar, hal ini mengidikasikan tingkat pencemaran bakteri pada makanan dan minuman jalanan masih tinggi.
