    Rubber plant (Hevea brasiliensis (Willd. ex A. Juss) Mull. Arg.) is one of commercial commodities in Riau Province that distributed at several plantation centers such as Bengkalis Regency, Kampar, Kuantan Singingi, Meranti, and Rokan Hulu. The propagation of rubber plants in long period of time causes variety in latex production. Nowdays, most of the rubber plantations in Riau Province use unstandardized seedling that result in diverse and low latex production. The first way to increate latex production is to identify the morphological characters related to latex production. This research was aimed to find out the characters that had correlation with latex productivity level. This research used exploration method. Samples such branches, leaves, and seeds from 10 rubber trees for each regency were collected, hence there were 50 samples were analyzed. The sample collection was based on the production level. A total of 66 morphological characters were analyzed. Seven out of 66 observed character had a correlation with latex production i.e. dense branch of tree, dense leaf of tree, dense leaf of branch, length petiole of main leaf, length petiole of leaflet, length of leaf, and wide of lamina. These characters can be used to select superior rubber plant.

    Celery is one of the most famous vegetables in the world because it has high economic value and useful as medicine. Celery can grow well in the highland with cool climate. The problem which arise the most is lack of interest of vegetable farmer to plant celery in the lowland because usually celery will grow stunted in incompatible environment. Informations about celery growth in lowland and hot climate are necessary to determine the best treatment which helps celery growth in lowland. The aims of this study was to determine type and the best liquid waste concentration for celery growth in Pekanbaru. Liquid waste used was bamboo shoots and jengkol skin. Celery seeds were obtained from farm store in Pekanbaru then planted in Botanical Research station FMIPA UR. Parameters observed were root length and volume. This study was carried out using completely randomized design (CRD) with 10 treatments repeated 4 times. Data was analyzed by ANOVA followed by Duncan's Multiple Range Test at 5%. Applications liquid wastes either alone or in combination showed significant effect on roots length, and roots volume. The result showed that 6,0 cc (R2) liquid of bamboo shoot gave the best effect on all parameters

    DAYA TUMBUH KECAMBAH DAN WAKTU BERTUNAS KACANG HIJAU (Vigna radiata L.) GENERASI F3

    The improvement program of mung bean (Vigna radiata .L) can be done by crossbreeding, selection, and testing of the progeny to obtain new varieties. This study aims to analyze the growth ability and germination time of mung bean sprout, F3 generation. The study was conducted at the experimental station of Biology, Faculty of Math and Natural Science, University of Riau from January to April 2015. A total of 693 mung beans seeds (F2 generation) were planted and observed. The results showed that the skin color of mung beans pods (F3 generation) have a variation. The grouts ability of mung bean sprout growth ability (72,22%) was F2-13 while the lowest (11,90%) is shown by F2-15. In Kenari and Kampar parent, the percentage of sprout growth ability were 61,9% and 57,14%, respectively. The bud forming time of mung bean (F3 generation) ranged from 5,60 to 6,83 days. The fastest forming time was found in F2-14 plot with 5,6 days in average. While the slowest time of forming bud was found in F2-12 plot with an average of 6,8 days. The germination time of Kenari and Kampar sprout were 6,17 and 6,12, respectively.

    DESKRIPSI KARAKTER MORFOLOGI UBI KAYU PUCUK HIJAU (Manihot essculenta Crantz) ASAL TALUK KUANTAN

    Cassava (Manihot esculenta crantz) is one of the agricultural commodity that produce tuber for food source in Indonesia. Cassava plants can be developed through plant breeding program that require plasma nuftah with high genetic diversity. This study aimed to describe the morphological characters of “Pucuk Hijau” cassava from Taluk Kuantan. This research was conducted from September 2013 to June 2014 in the garden of Biology ,Faculty of Mathematic and Natural Science University of Riau. Materials used in this study were “Pucuk Hijau” cassava seeds and cow manure. The procedure involves planting and observation of morphological characters. The results showed that the morphological characters of cassava shoots are, green and un-grove stem surface, long ptiole (20 cm), conical-cylidrical tuber shape, light brown outer tuber, thin (1 mm) and purple tuber, with white inner tuber.

    Eksplorasi Penanda Genetik Spesifik Ikan Endemik Sungai Paparan Banjir Riau (GENUS Kryptopterus DAN Ompok) Berbasis Gen Cox-1 Dan D Loop Region

    Sungai paparan banjir atau floodplain river di Indonesia hanya ditemukan di Kalimantan, Palembang, Jambi dan Riau. Sungai paparan banjir di Riau umumnya dicirikan oleh warna perairan coklat tua, transparansi tinggi dan pH relatif lebih rendah. Ada beberapa jenis ikan yang bersifat endemik di sungai paparan banjir yaitu jenis-jenis ikan dari genus Kryptopterus dan Ompok yang termasuk famili Siluridae yang dikenal secara umum di Indonesia sebagai ikan lais, dan secara spesifik di Riau dikenal dengan nama ikan selais. Ikan selais merupakan maskot kota Pekanbaru dan ikan konsumsi yang bernilai ekonomis tinggi. Oleh karena potensinya, eksplorasi informasi yang mendalam mengenai ikan ini sangat perlu diketahui dengan pasti terutama dengan teknik molekuler yang telah berkembang pesat akhir-akhir ini. Sampai saat ini, informasi ilmiah mengenai penanda genetik ikan endemik sungai paparan banjir khususnya dari provinsi Riau belum begitu banyak diketahui. Penelitian yang telah dilakukan umumnya masih terbatas secara morfologi. Penelitian ini bertujuan untuk menentukan penanda genetik spesifik ikan endemik sungai paparan banjir Provinsi Riau, berbasis gen penyandi protein yaitu gen Cox-1 pada tahun pertama, sedangkan pada tahun kedua menentukan penanda genetik spesifik berbasis daerah bukan penyandi protein (non coding region) yaitu D-loop region. Penelitian mengenai penanda genetik spesifik dapat digunakan sebagai DNA barcode yang mencirikan identitas jenis dan asal-usul jenis tersebut. Dalam penelitian tahun kedua, setiap individu dari masing-masing jenis yang didapatkan dari genus Kryptopterus dan Ompok yang menjadi objek penelitian, dilakukan isolasi DNA total dari bagian ototnya. Daerah D-loop region DNA mitokondria dari DNA total akan diamplifikasi dengan primer forward dan reverse menggunakan mesin PCR. Kualitas hasil amplifikasi dapat dilihat pada gel agarose 1,2 % menggunakan mesin elektroforesis. Daerah D-loop region yang teramplifikasi dilakukan perunutan DNAnya dengan mesin perunutan (seqencer) DNA. Hasil runutan yang terdiri dari basa nukleotida DNA mitokondria dianalisis dengan program Mega versi 4,0 untuk mendapatkan data penanda genetik spesifik sebagai DNA barcode masing-masing jenis dan DNA barcode lokasi asalnya berbasis D-loop region.

    LBKURCC146 and LBKURCC147 are endophytic bacteria that isolated from Harupat ferns stem in Pekanbaru, Riau that resistant to acidic pH. The purpose of this study was to extract the DNA of endophytic bacteria Enterobacter cloacae LBKURCC146 and LBKURCC147. DNA of LBKURCC146 and LBKURCC147 bacteria were isolated after culturing for 24 hours inccubation using Geneaid Kit. The size of bacterial DNA obtained from the isolation results is above 10037 pb. DNA can be used for further step such as amplification (PCR).

    Riau Province has the typical of floodplain river. One of the fish that live in this river ecosystem is Kryptopterus limpok. Information of molecular barcodes of mitochondrial cytochrome c oxydase subunit 3 (COX3) gene from K. limpok is still unknown. The objective of this research was to obtain the DNA fragment of the mitochondrial COX3 gene K. limpok from Kampar River, Tapung River, and Indragiri Hulu River. Total DNA isolated were taken from the muscle of fish-tail. The total DNA was electrophoresed on 1,2% agarose gel and then visualized using UV transiluminator (WiseUv WUV-M20, Daihan Scientific). PCR amplification was performed using a primer pair of COXIIIF1 (forward) 5’GCT TAG GTT TCA CCA CAA GA3’ and COXIIIR1 (reverse) 5'CGA ACA CAA ATC AGT GGT GTT CT3'. PCR condition based on Elvyra dan Duryadi (2007) with modification. Total DNA concentrations produced ranging from ±300 ng/μl and the fragment length of the mitochondrial COX3 gene K. limpok was 660 bp

    Padi (Oryza sdtiva L.) rnerupakan sumber pangan bagi masyarakat Indonesia yang dikonsumsi tidak kuran g dari 200 juta penduduk. Konsumsi beras rata-rata 133 kglkapita/tahun, maka total kebutuhan beras 26,6 juta ton/tahun (Husodo 2007). Pertarnbahan jurnlah penduduk nrendorong rneningkatnya kebutuhan akan beras, oleh karena itu perlu digalakkan usaha peningkatan produksi beras untuk memenuhi kebutuhan tersebut. Angka Tetap (ATAP) prodtrksi padi tahun 2010 sebesar 574.864 ton padi Gabah Kering Giling (GKG) atau meningkat dibanding produksi padi pada tahun 2005 sebesat 425.095 toMra (BPS 2011). Peningkatan produksi padi tersebut belnm mencuktrpi kebutuhan konsumsi beras penduduk R.iau sebesar 659.6rc tonlha. Hal ini menyebabkan tingginya ketergantungan pasokan beras dari Provinsi lain seperti Sumatera Barat atau Sumatera Utara. Untuk mengurangi ketergantuttgaf tersebut, pemerintah Provinsi beserta pelnerintah Kabupaten se Riau mengeluarkan kebijakan peningkatan produksi beras. Kebiikan tersebut dapat diwujudkan dalam bentuk progrimr ekstensifikasi manpun intensifikasi. Upaya ek-stensiJikasl dilakukan dengan rnengernbangkan areal pertanian seperti mengembangkan varietas padi pada lahan gambut yang barryak terdapat di Provinsi Riau. Usaha inten.sifikasr dilakukan dengan cara perbaikan varietas padi melalui pengembangan galur-galur yang mempunyai potensi produksi tinggi dengan biayaminimal.

    Identifica'i'ion Of Period Needed Each Phase In Pyrrosia lanceolata (L.) Farwell Fern Root Cells During Mitosis

    The objectives of this research were to identification of period needed each phase in Pyrrosia lanceolata (L.) Farwell fern root cells during mitosis. Pyrrosia lanceolata (L) farwell ferns were obtained from Pekanbaru, Riau. Each phase in mitosis observed by made root cells fresh specimen by squash method. We found that between 3.20 am. Until 3.35 am, overall phase could observe. This result showed that Pyrrosia lanceolata (L._) farwell fern root cells active mitosis at that period or time. Prophase and telophase would be seen at every specimen which obtained from different time. In contrast, it was difficulty get anaphase and metaphase. Therefore, we could not now make some conclusion about period needed each phase in Pyrosia lanceolale (L.) Farwell fern root cells during mitosis.

    ISOLASI DAN AMPLIFIKASI FRAGMEN GEN COX3 PADA IKAN Ompok hypophthalmus (Bleeker 1846) DARI SUNGAI TAPUNG PROVINSI RIAU

    Selais danau fish (Ompok hypophthalmus) is one of the fish which inhabit the floodplain river. Genetic information on Ompok hypophthalmus is still limited. This research aims to obtain total DNA of O. hypophthalmus and DNA fragment amplified from COX3 gene. Samples of ten O. hypophthalmus, retrieved from a floodplain river, Tapung River, were isolated using the isolation Kit. Purification of DNA has been done using Dneasy Blood and Tissue Kit from Qiagen. Total DNA was amplified using COX3 F1 and COX3 R1 primers which were designed using sequence of COX3 gene from Ictalurus punctatus (Genbank 2002). The result indicated that eight samples of total DNA were successfully isolated from ten of fish samples taken. Amplification of total DNA using primer COX3 F1 and COX3 R1 produces DNA fragments which sized 671 bp.

    Cassava (Manihot esculenta Cranzt.) is a species that has high adaptability to acidic and poor nutrient soil in Riau Province. This province has high cassava genotypes that distributed in several districts, including Rokan Hilir. The variegata and keriting cassava are cassava genotypes from Rokan Hilir, but the genetic studies to isolation DNA have never been conducted. This study was aimed to isolate and electrophoresis the total DNA of variegata dan keriting cassava. The research methods included the isolation of DNA from fresh leaves of the variegata dan keriting cassava using CTAB (Cetyl Trimethyl Ammonium Bromide) method and the electrophoresis was conducted ina 1,2% agarose gel to know the quantity and quality of total DNA obtained. The results showed that the quality of the total DNA of variegata dan keriting cassava with thick band and intact. The concentration of total DNA obtained approximately 700-1000 ng/ μl

The maman plant is a traditional plant of the Rokan Malay tribe which is often used as traditional medicine and can be processed into a special food called “maman orange” which is commonly found in Rokan Hulu, Rokan Hilir and Kampar Kiri Regencies, Riau Province. This study aims to extract the total DNA of plants. Research methods include total DNA isolation and electrophoresis. The resulting total DNA has a thick, bright band, has length of more than 10,000 bp.

    Pandan plant from Kubang is a plant of the Pandanaceae family. Morphologically, pandan from Kubang is similar to Benstonea sp from Pelalawan, so it is necessary to analyze molecularly. The first step in molecular analysis is isolation of genomic DNA. This study aimed to isolate total DNA of pandan from Kubang. Research method include total DNA isolation and electrophoresis. Result of total DNA extraction showed a thick and single band with length of more than 10,000 bp.

    Selais Kaporeh (Ompok eugeneiatus) is an endemic fish in floodplain river. The genetic information of Ompok eugeneiatus is still limited. This research aims to isolate and amplify Cytochrome c Oxydase sub unit 3 (COX3) gene from Ompok eugeneiatus originated from Kampar River. This study was conducted from November 2014 to April 2015. Cytochrome c Oxydase sub unit 3 gene was amplified using COX3-F1 and COX3-R1 primers which were design from COX3 gene of Ictalurus punctatus (GenBank 2004). PCR condition based on Elvyra and Duryadi (2007) with modification. The result indicated that COX3-F1 and COX3-R1 primers can be used to amplify COX3 gene of Ompok eugeneiatus with size of 642 bp.

    Lopang plants are annual herbaceous plants that have been cultivated in Riau Province. This plant grows creeping and dominantly growing in open land which is used as a fields. This plant is used as a vegetable and is believed to be an herbal medicine for certain diseases by Riau people. This study aims to extraction the total DNA of Lopang plants. Sour eggplant is one of important ingredients in malay cuisines, especially in Siak and Kampar, such as “asam pedas ikan tapah”. This study aimed to isolate sour eggplant DNA using Genomic Mini Kit Plant. Research methods consisted of isolation of genomic DNA and electrophoresis.     Endophytic bacteria are living and saprophyte bacteria associated with plant tissue without causing harm to the plant. One of endophytic bacteria is from the genus of Bacillus sp. Molecular analysis using 16S rRNA genes needs to be done to identify and determine the species of endophytic bacteria. The purpose of this study was to isolate DNA of endophytic bacteria originated from palm oil plants by using 16S rRNA gene. This research was conducted from February to May 2016. Bacteria was isolated from 3 healthy palm plants, existed around oil palm crops which were afflicted by disease of rotting base on the stem. Of each plant, bacteria were isolated from the stem, roots, stems and leaf. Research methods included the manufacture of medium Nutrient Agar (NA) and Nutrient Broth (NB), rejuvenation of the isolates of bacteria, bacterial DNA isolation and electrophoresis. The whole (12) isolates of endophytic bacteria which successfully isolated showed that the total DNA retrieved was intact, characterized by a thick and intact band

    Pucuk seminyak or siminyak is a perennial plants that are used by people, especially the traditional Rokan Malay people, to be made into a delicious food. Pucuk seminyak can be found in open or shaded areas in forests or plantations. This study aimed to isolate total DNA from pucuk seminyak with sample codes PS1 and PS2 from Kampar Regency. This research extracted total DNA using the Genomic DNA Mini Kit Plant (Geneaid) and electrophoresis with 1% agarose gel. The visualization results of total DNA bands from the pucuk seminyak in this study showed that the bands obtained were thin and smears with size of more than 10.000 bp.

    Kabau (Archidendron microcarpum) is an uncultivated plant that belongs to the Fabacea family. Kabau plants have potential as medicinal plants because they contain lectins as antibacterial, antidiabetic, antifungal, and antidiuretic agents. Molecular information about kabau plants in the Genbank database is not yet available, therefore this research is carried out to isolate total DNA from kabau plants.. Research methods included isolation of total kabau plant DNA samples K3 and K4 from fresh leaves using the Genomic DNA Mini Kit Plant (Geneaid) DNA isolation kit and electrophoresis using 1% agarose gel to check the quality and quantity of total DNA. The results showed that the kabau plant DNA bands K3 and K4 samples were intact, bright, single, and not smeared with a size of more than 10,000 bp.
